Digital PCR
Partitioning & Multiplex Amplification

This is where the biochemistry turns into digital data. The sample mixture is loaded directly into the digital PCR system, whether it utilizes microfluidic nanoplates, chips, or droplet generation.

  • Micro-Partitioning: The sample is instantly distributed across thousands of isolated nanoliter cavities or oil droplets. Individual targets and their bound DNA-labeled antibodies are locked into separate, independent micro-reactions.
  • Proximity-Driven Amplification: Once partitioned, the standard dPCR master mix amplifies the DNA barcodes. A distinct fluorescent signal is generated only if the specific target markers are present together in that individual partition.
  • Multiplexing Opportunities: By using the multiple fluorescent dye channels on your dPCR hardware, you can simultaneously detect different target combinations, subpopulation markers, or distinct protein complexes in a single run.
  • Direct Molecular Counting: Because dPCR operates on a binary (positive/negative) readout per partition, the system counts the absolute number of fluorescent events. No reference standards or standard curves required.

PICO Amplification Core Kit

The PICO Amplification Core Kit enables the detection and absolute quantification of Extracellular vesicles, proteins, protein interactions, and post-translational modifications. The workflow includes the lysis of the sample, followed by the incubation of lysates with the labeled antibodies. After the incubation the sample is highly diluted and loaded into a dPCR plate. During the dPCR reaction, the PICOglue Labels are amplified and fluorescent signal is generated and recorded. After the dPCR run, the raw data is uploaded and analyzed with our online software.

The AMC-Kit contains materials to perform 120 dPCR reactions in 24-well dPCR plates or 480 dPCR reactions in 96-well dPCR plates.

We offer a professional service for labeling antibodies of your choice with PICOglue Labels, providing a highly sensitive and specific probe for various PICO applications. 

Our service uses the PICOglue Labeling Technology to ensure the superior quality and reliability of the labeled antibodies. We guarantee at least 75% labeling efficiency (many times close to 100%), with a labeled antibody stability of at least 12 months. The yield is high and the amount of labeled antibodies are enough for thousands of PICO assays. The expected concentration of the labeled antibody is in the range of 5 × e10 - 5 × e12 cp/µL (copies per microliter).

The PICO EV Q Kit enables the absolute quantification and characterization of Extracellular vesicles based on their surface markers. 
The workflow includes the incubation of your purified EVs with labeled antibodies. After incubation, the sample is diluted and loaded onto a dPCR plate. During the dPCR reaction, the PICOglue Labels are amplified, generating a fluorescent signal that is recorded. After the
dPCR run, the raw data is uploaded and analyzed with our online software.

The PICO EV Q Kit contains materials to perform 150 dPCR reactions in 24-well dPCR plates or 600 dPCR reactions in 96-well dPCR plates.
