Key capabilities and possibilities using PIQuant:

  • PICO Experiment Data Analysis: The primary purpose is to analyze data from a Protein Interaction Coupling (PICO) immunoassay experiment, which is used for the characterization and quantification of Extracellular Vesicles (EVs), proteins, protein interactions, and post-translational modifications.

  • Quantification Options: It offers two main evaluation options for PICO data:
    • Relative Quantification (RQ): Uses an external standard or control sample for internal reference.
    • Absolute Quantification (AQ): Analysis performed without an external standard.
  • Data Processing: It allows users to select content to analyze (antibody mix, antibody pairs, samples) and how the couplex data will be processed. Recommended processing for maximal accuracy of Absolute Quantitative data includes:
    • Lambda normalization, Antibody Binding Control (ABC), and Labeling Efficiency (LE) corrections (Couplexes LN-ABC-LE).
    • Note: The labeling efficiency correction should not be applied to Extracellular Vesicle (EV) samples.
  • Quality Control: It features a Quality Control tab that details performance features such as the number of valid partitions, positive non-template control (NTC) partitions, average lambda, and ABC values, with acceptable ranges suggested by PIQuant.

  • Evaluation and Results: The Evaluation tab displays experimental results, including:
    • Couplex graphs that give a first insight into the relative abundance of the target.
    • The ability to run absolute quantification after indicating antibody concentration and target lambda (e.g., 500 pM and 0.15 for regular AQ).
    • The option to select only data from the "high-side" or "low-side" for absolute quantification, which is necessary due to the "hook effect" that can occur in immunoassays.
  • Data Download: Users can download the analysis data and graphs once the analysis is completed.

  • Device Support: PIQuant currently supports dPCR devices from Qiagen and Bio-Rad (beta).
