Quantification of protein-protein interactions (PPI) with PICO

In this application note, we used the PICO technology to detect and measure HER2 and HER3, as well as their interaction. With Absolute Quantification, we demonstrated that PICO is about 200 times more sensitive than co-immunoprecipitation (coIP). This makes PICO a powerful solution for quantifying protein interactions and monitoring signaling pathway activity.

Quantification of post-translational modification (PTM) with PICO

In this application note, we demonstrate how the PICO technology can be used to both detect and accurately quantify the protein 4EBP1 and its phosphorylated form. By applying an Absolute Quantification approach, we measured the precise amount of phosphorylated 4EBP1 in BT474 and MCF7 cells. When these cells were treated with the kinase inhibitor Dactolisib, the phosphorylation levels of 4EBP1 were reduced by approximately 200-fold, clearly illustrating the inhibitor's impact on this signaling pathway.

Quantification of extracellular vesicles (EVs) with PICO

In this application note, we detected and quantified extracellular vesicles (EVs) from serum or cell supernatant using the PICO technology. The EVs were measured with two different detection strategies: 1) using a CD63 antibody, halved and labeled with two different PICOglue Labels, we quantified CD63+ EVs, while 2) using CD63 and CD9 antibodies, we quantified CD9/CD63+ EVs. These findings highlight the versatility of PICO in the sensitive and accurate detection of various protein targets.

Comparing the Sensitivity of PICO vs. Western Blot

In this application note, we compared the sensitivity of PICO to Western blot. We chose to detect the breast cancer biomarker HER2/ErbB2 in two different breast cancer cell lines, BT474 and MCF7. Through our analysis, we found that PICO demonstrated approximately 200-fold higher sensitivity compared to western blot. These findings further support the advantages of using PICO for sensitive and accurate protein detection.

Absolute vs. Relative Quantification with PICO

In this application note, we compared the two PICO quantification methods: the Absolute Quantification and the Relative Quantification. Using HER2 as the model protein, we showed that BT474 cells have approximately 120-fold more HER2 than MCF7 cells.
